132 Women Over 40: Oral + Topical Resveratrol Cut Wrinkles in 8 Weeks

French ingredient supplier Lallemand Bio-Ingredients conducted a double-blind, randomized, placebo-controlled trial in 132 women aged 40+ combining oral and topical trans-resveratrol. The result, published in Frontiers in Aging, showed significant wrinkle reduction and normalized sebum compared to placebo.

Four-Group Design

Participants were randomized into four arms:

  • Oral 75 mg (1 capsule/day) + Topical 1.5% (1 g cream twice daily)
  • Oral only
  • Topical only
  • Placebo

After 8 weeks, the oral + topical combination group showed the largest wrinkle score reduction. Both single-route groups improved versus placebo, but combination outperformed either alone.

How Resveratrol Works

Resveratrol is a polyphenol naturally present in grape skins, red wine, and peanuts. Three main mechanisms:

  1. Sirtuin (SIRT1) activation: longevity-related enzymes that regulate DNA repair, mitochondrial function, and stress response. NAD+ dependent.
  2. Antioxidant action: neutralizes reactive oxygen species (ROS) to defend against UV-induced skin damage.
  3. Collagen support: inhibits matrix metalloproteinase (MMP) activity that breaks down collagen.

First Single-Ingredient Clinical Evidence

Notably, this trial is the first to establish resveratrol as a single active for skin aging. Most prior resveratrol research used combination formulations or was limited to animal studies. Confirming single-ingredient effect at 132-person scale with placebo control moved resveratrol’s scientific position up a level.

Wine Isn’t Enough

The “resveratrol in red wine” claim is famous, but practically a glass of wine (150 ml) contains only 0.2-2 mg. Matching the trial’s 75 mg requires drinking dozens of glasses daily, far outweighed by alcohol harm. Concentrated supplements at 75-150 mg/day are the practical delivery.

What Sebum Normalization Means

The trial’s reporting of sebum normalization is notable. After 40, estrogen decline reduces facial sebum production, which contributes to dryness, loss of elasticity, and pruritus. If resveratrol restores sebum to normal levels, it complements wrinkle reduction by supporting barrier function itself, not just surface smoothing.

Resveratrol and NAD+

Resveratrol is a sirtuin-activating compound (STAC). Sirtuins consume NAD+ while performing DNA repair, protein folding, and inflammation control. Resveratrol stimulates the enzyme; NAD+ (or its precursors NR and NMN) supplies fuel.

This relationship has made resveratrol + NAD+ precursor combinations a new area of interest in 2026 skin longevity routines.

Practical Guide

  • Oral dose: 75-150 mg/day (trans-resveratrol)
  • Topical concentration: 0.5-2%
  • Timing: with breakfast (fat-soluble)
  • Pairing: vitamin C, vitamin E, and NAD+ precursors compound the effect
  • Caution: consult clinician if on anticoagulants

Resveratrol has been marketed as “the miracle wine molecule” for decades, with limited single-ingredient clinical evidence. Lallemand’s 132-person, 8-week trial closed that gap, positioning resveratrol as a standard ingredient in skin longevity routines.
